چکیده

Low concentrations of the antibiotic griseofulvin were found to cause increased frequencies of somatic segregation due to chromosome nondisjunction in a diploid strain of Aspergillus nidulans.

Induced Somatic Segregation in Aspergillus Nidulans.

ETEROZYGOUS diploid strains of Aspergillus nidulans spontaneously proHduce segregant clones belonging to three classes: diploid segregants which have undergone mitotic crossing over; haploid segregants with random redistribution of whole chromosomes (PONTECORVO, TARR GLOOR and FORBES 1954) ; and "nondisjunctional" segregants with all the markers on both branches of one chromosome in the homozyg...

Analysis of Vegetative Segregation and Recombination in Penicillium Chrysogenum.

N the basis of an analysis of vegetative segregation in heterozygous diploids of 0 Aspergillus nidulans, PONTECORVO et al. (PONTECORVO 1953 ; PONTECORVO, TARR GLOOR and FORBES 1954) concluded that the segregant nuclei arise as a consequence of two independent processes. The first is mitotic crossing over (STERN 1936), giving rise to diploid nuclei which are homozygous for some of the markers. T...

Mitosis in Aspergillus nidulans.

Mitosis in growing hyphae of haploid and diploid Aspergillus nidulans has been studied with the light microscope in Helly-fixed preparations. At the periphery of the nucleus a sharply defined granule is invariably found which has strong affinity for acid fuchsin. Mitosis begins with the duplication of this granule and the development inside the nucleus of a gradually lengthening fibre between t...

An extragenic suppressor of the mitosis-defective bimD6 mutation of Aspergillus nidulans codes for a chromosome scaffold protein.

We previously identified a gene, bimD, that functions in chromosome segregation and contains sequences suggesting that it may be a DNA-binding protein. Two conditionally lethal mutations in bimD arrest with aberrant mitotic spindles at restrictive temperature. These spindles have one-third the normal number of microtubules, and the chromosomes never attach to the remaining microtubules. For thi...

Down-Regulation of sidB Gene by Use of RNA Interference in Aspergillus nidulans

Background: Introduction of the RNA interference (RNAi) machinery has guided the researchers to discover the function of essential vital or virulence factor genes in the microorganisms such as fungi. In the filamentous fungus Aspergillus nidulans, the gene sidB plays an essential role in septation, conidiation and vegetative hyphal growth. In the present study, we benefited from the RNAi strate...
